NEW YORK--Enzeco® Trypsin/Chymotrypsin 1:1 is a new amorphous powder derived from North American bovine pancreas from Enzyme Development. BothTrypsin, 3.4.21.4, and Chymotrypsin, 3.4.21.1, are classed as serine proteases. Chymotrypsin preferentially cleavestryosine, tryptophan, phenylalanine and leucine. The pH optimum is 8 to 9 with a temperature optimum of 35 C. Trypsin preferentially cleaves arginine and lysine, and its pH optimum is 8 to 9 with a temperature optimum of 45 C. The activity of the enzyme is determined by the method published in the U.S. Pharmacopoeia. Trypsin activity is approximately 1,000 units/mg. A unit of activity is defined as the amount of enzyme causing an increase in absorbance at 253 nm of .003 per minute at 25 C. Chymotrypsin activity is approximately 1,000 units/mg. A unit of activity is defined as the amount of enzyme causing a decrease in absorbance at 237 nm of .0075 per minute at 25 C.
